چکیده
Üç alfa-karbonik anhidraz, insan karbonik anhidraz I, II (hCA I, II) ve koyun böbrek karbonik anhidraz (sCA) saflaştırıldı ve enzimlerle oksitetrasiklinin kinetik etkileşmeleri incelendi. Saflaştırma prosedürü homojenat hazırlama ve Sepharose 4B-trizonsüllfanilamid afinite kramatografisinden oluştu. Oksitetrasiklin hCA I, II ve sCA enzim aktivitelerinde in vitro aktivatör etkisi gösterdi. Bu antibiyotik için % aktivite-[A] grafiği kullanılarak % aktivite değerleri belirlendi.

Bacterial Carbonic Anhydrases as Drug Targets: Toward Novel Antibiotics?
Carbonic anhydrases (CAs, EC 4.2.1.1) are metalloenzymes which catalyze the hydration of carbon dioxide to bicarbonate and protons. Many pathogenic bacteria encode such enzymes belonging to the α-, β-, and/or γ-CA families. In the last decade, the α-CAs from Neisseria spp. and Helicobacter pylori as well as the β-class enzymes from Escherichia coli, H. pylori,Mycobacterium tuberculosis, Brucell...
متن کاملAn Overview of the Bacterial Carbonic Anhydrases
Bacteria encode carbonic anhydrases (CAs, EC 4.2.1.1) belonging to three different genetic families, the α-, β-, and γ-classes. By equilibrating CO₂ and bicarbonate, these metalloenzymes interfere with pH regulation and other crucial physiological processes of these organisms. The detailed investigations of many such enzymes from pathogenic and non-pathogenic bacteria afford the opportunity to ...

Citation: Aggarwal M, McKenna R. Carbonic Anhydrases: Nature’s Way to Balance CO2 Concentration. Biochem Mol Biol J. 2016, 1:1. The carbonic anhydrases (CAs; EC 4.2.1.1) are a family of structurally diverse (in both fold and oligomeric state), yet efficient metalloenzymes that catalyze the reversible hydration of CO2 and bicarbonate. They are categorized into five distinct classes (α, β, γ, δ, ...
متن کاملHuman Carbonic Anhydrases II. SOME PHYSlCOCHEMICAL PROPERTIES OF NATIVE ISOZYMES AND OF SIMILAR ISOZYMES
Detailed studies of a relatively large number of human erythrocyte carbonic anhydrases of different electrophoretic mobilities have shown that individually each is very similar to one of the major isozymes, either B or C. The enzymatic activities, the zinc, nitrogen, and amino acid contents, and the molecular weights of members of a given type are similar. Some differences in the tryptic peptid...
متن کاملCarbonic Anhydrases in Cnidarians: Novel Perspectives from the Octocorallian Corallium rubrum
Although the ability to elaborate calcium carbonate biominerals was apparently gained independently during animal evolution, members of the alpha carbonic anhydrases (α-CAs) family, which catalyze the interconversion of CO2 into HCO3-, are involved in the biomineralization process across metazoans. In the Mediterranean red coral Corallium rubrum, inhibition studies suggest an essential role of ...
